Crafting;
One of the easiest way to obtain the reagent is by transmuting it using the Transmute Nightshade Recipe available from Wul'yahm, the Apprentice Crafter in The Oasis, Krokotopia. She sells the recipe for 200 gold.
The recipe allows you to convert 15 Deep Mushrooms to one Nightshade.
Harvest;
Nightshade can be found as a Rare Harvest from Deep Mushrooms found practically all over the spiral, so keep a look out.
Crowns;
Also, a 100 Deep Mushroom Bundle is available in the Crown Shop for about 599 Crowns or so if I recall correctly.
Assuming it's the first, Lord Nightshade is found in the Haunted Cave, Wizard City.
Being the final Storyline boss in Wizard City, you must complete all three streets (Cyclops Lane, Triton Avenue & Firecat Alley) in addition to beating Foulgaze, another boss found in OdleTown to finally have access to his tower at the end of the Haunted Cave.
The only other way is to port to a friend on the inside.
You get access to Nightside upon gearing up to face Nightshade.
