I saw an item that had a -100% mana stat on it, does this mean that I will lose 100% of my mana?

It means while wearing that gear you will have 0 mana, When you remove that gear it will show you have 0 mana, thus you must collect wisps, play mini-games, or level up to have mana again.

it means the item will take away one hundred points of mana for example:

if i have 246 mana and i equip the item you describe i will be left with 146 mana points

It states that it takes 100% not 100 mana so,...what would be the point of using such an item if it removes all your mana or even removes a good chunk of it which you worked hard to get up?

PVP doesn't require mana
so the gear in question are actually pvp gear

whatever the gear you saw i'm positive it was some sort of PvP gear, no? well that's because you don't need mana for PvP! it's KI's way of limiting that gear to the arena.
